Arylethynyl nucleosides derived from fluorescent polycyclic aromatic hydrocarbons, e.g. their first representative 5-(1-pyreneethynyl)-2’-deoxy-uridine 1,1’2 are both useful fluorescent labels and prospective broad spectrum antivirals. Arylbutadiynyl nucleosides are much less studied. We reported the synthesis of 5-(1-pyrenebutadiynyl)-2’-deoxy-uridine 2;2 Brown and coworkers described incorporation ofanthracenebutadiynyl nucleoside 3 into oligonucleotides as base discriminating fluorescent label.7 Recently, other (non-polycyclic) 5-(R-butadiynyl)-2’-deoxy-uridines showed medium activity against vesicular stomatitis virus. All the data prompted us to prepare 2- and 4-pyrenebutadiynyl nucleosides 4 and 5 (Fig. 1) for fluorescent and antiviral studies.
