Arabidopsis thaliana mutant lel demonstrate the loss of petals and stamens, male sterility and severe abnormalities in development. We suggest involvement of gene LEL in maintenance of floral meristem. We analyzed the double mutants lel wus-1, lel syd-1, lel clv3-2, lel er to reveal genetic interactions between LEL and these genes participated in control of meristem development. Our results supposed that LEL is a negative regulator of apical meristem and positive a regulator of floral meristem. We use RNA-seq in two independent repeats to compare expression level of genes in wild-type A. thaliana and lel. We found about 1500 differentially expressed genes. Among them there were main stem cell activity regulator WUSCHEL which expression decreased five times, its negative regulator CLAVATA1 increased by half and other genes taken part in control of flower development and transition to flowering. Data of RNA-seq were justified with RT-qPCR experiment. The results of RNA-seq agree with mutant analysis, so we define LEL as novel regulator of meristem.
