One of the most commonly exploited oncotherapeutic strategies is based on the use of special chemical agents which are able to induce DNA damage and thus trigger apoptotic death of cancer cells. However, these widely used drugs have significant toxic effects because normal cells also undergo apoptosis after treatment. Hence one of the most important goals in medicine is the search and investigation of different strategies to enhance the efficiency of chemotherapeutic agents. Calorie restriction was shown to protect normal cells and sensitize cancer cells to chemotherapeutic agents. Different susceptibility of normal and cancer cells to stress stimuli upon calorie restriction is called different stress resistance. This could allow decreasing the dose of chemotherapeutic drugs and reducing their toxicity. Among different variants of calorie restriction short-term calorie restriction was shown to be the most promising approach for enhancement of chemotherapeutic efficiency. In our studies we examined the mechanisms how serum deprivation, which serves as an in vitro model of CR, modulates apoptosis in two cancer cell lines (ovarian carcinoma line Caov-4 and cervical adenocarcinoma line HeLa) in response to treatment with DNA-damaging agent cisplatin.
