Elevation of background radiation level due to various technogenic factors and the problem of cosmic radiation, which impedes the prolonged stay of organisms in space, aim to study new mechanisms for increasing innate radioresistance. The unique protein Dsup found in Tardigrade is a nuclear localized protein with radioprotective properties that was confirmed in experiments with transgenic plants and cell cultures. The main predicted mechanism of Dsup action is its binding with nucleosomes that causes shielding DNA from ROS that decreases DNA damage and increases the cell survival. However, the issue whether making a kind of additional isolation around DNA and hindrances for molecular movement induced by Dsup-nucleosome complex influence on transcription and reparation processes is unclear. In our work we generated several D. melanogaster lines constitutively expressing Dsup under Act5C gene promoter. Transcriptome analysis with GeneChip Drosophila Genome 2.0 Arrays for control initial y1w1118 line and Dsup expressing lines revealed changes in the expression of genes involved in transcription, chromatin silencing, chromosome organization, that may indicate disorder traces in DNA-RNA machinery or chromatin compaction. Moreover, in Dsup expressing lines we observed alteration in expression of genes concerning immune response, which may indicate that flies expressing Dsup were feeling some stress. Dsupexpressing and control lines were irradiated with gamma rays at a dose 1000 Gy and we observed the increasing of survival rate in the first three days after irradiation in Dsup expressing flies. The intention of this work is to perform transcriptome analysis of irradiated and nonirradiated control and Dsup expressing lines and estimation of lifespan and locomotion in them.
