Neuropathy target esterase (NTE) ia a specific target for neurophathic organophosphates (OP) and it seems to be a site of initiation of OP-induced delayed neuropathy (OPIDN). NTE inhibition within hours of exposure to OP predicts potential for the development of OPIDN. Fast and effective methods monitoring of neuropathic OP exposure to humans are highly required. New biosensor method for the analysis of NTE and its inhibitors is based on the combination of NTE enzymatic hydrolysis of phenyl valerate with phenol detection by carbon-paste tyrosinase electrode. The use of the tyrosinase electrode leads to 10-fold improvement in the sensitivity of the analysis of NTE activity in comparison with the spectrophotometric detection. The tyrosinase electrode was found to be suitable for measurements of NTE activity in whole human blood when the application of the usual spectrophotometric detection is considerably limited. The results of inhibitor specificity obtained for mipafox and DFP titration of NTE in blood were close to those for neuronal NTE. NTE-like activity in blood was found to be equal 0.20 nmoles/min per mg of protein.
