Chromosome ends of all eukaryotes have nucleotides repeats (TTAGGG in vertebrates) protecting chromosomes from degradation as the result of incomplete replication of ends and also from end junction – telomeres. Telomeres participate in the cellular answer to DNA breaks. There is a protein complex that is bound to telomeres and consists of many proteins – shelterin, which components protect telomeres from recognition of systems of reparation and also attract telomerase, that elongate telomeres in gametes, stem cells and most of cancer cells. PAPR1 is a poly(ADP-ribose)-polymerase 1, which is a one of sensors to DNA breaks. It is known that overexpression of one protein of shelterin complex – TRF1 and also PARP1 is observed through patients with Duchenne muscular dystrophy that lead to telomeres shortening. Activity of some proteins of shelterin complex is regulated within PARylation. Telomeres and PARP1 play the important role in genome integrity maintenance. We obtained cell line with low expression of PARP1 gene and evaluated status and structure of telomeres and also telomerase functionating. The data received allow us to suppose that PARP1 regulates biogenesis of telomerase, namely assembly and stability of active enzyme and also makes telomeres more available to elongation.
