Целью данного проекта являются определение соединений с новыми молекулярными каркасами для противоопухолевой терапии.
Despite the significant number of anticancer drugs known to date, malignant tumors remain one of the main causes of death. In 2020, approximately 19.3 million new cases of cancer and almost 10 million deaths from cancer were recorded in the world. The most common types of cancer are lung and breast tumors. This project is aimed at finding new molecular scaffolds with antitumor potential against cells of these types of tumors, assessment of their mechanisms of action, and the creation of a database of structurally new biologically active compounds. For several groups of structurally related active compounds, basic data on cell-induced effects and molecular mechanisms of action will be obtained. The analysis of these data will allow us not only to select leader compounds, but also to construct structure-activity relationships for them for the targeted synthesis of next-generation antitumor compounds. To search for leader groups of molecules, about five thousand structurally new compounds and three thousand compounds selected in silico with predicted antitumor activity will be analyzed for cytotoxicity and selectivity of action against human cells of tumor and non-tumor origin. The data from the planned high-throughput screening of both collections, together with the results obtained during the preliminary studies, will lead to the creation of an original database of the effects of over ten thousand structurally diverse compounds on the selected human cell lines. We plan to identify few new chemical scaffolds (structural core for groups of active molecules) with high antitumor potential, by screening and optimizing the molecules selected in the screen and studying their mechanism of action.
