Аннотация:A fluorescence polarization immunoassay (FPIA) based on a polyclonal antibody for the determination of Sulfamethazine (SMZ) was developed. In order to optimize the FPIA procedures, three fluorescein-labeled, SMZ-FITC, sulfamerazine (SMR)-FITC and SMZ-dichlorotriazinylaminofluoreseein (DTAF) were synthesized and the influence of their structures on FPIA characteristics was studied. The optimized SMZ FPIA had a dynamic range from 5 to 458 mu g/L with IC50 value of 46 mu g/L and a detection limit of 1.6 mu g/L. The specificity of the FPIA for SMZ was investigated using other 16 sulfonamides and the cross-reactivity for SMZ, SMR and sulfamoxol (SMX) are 100%, 8.6% and 2.4%, respectively. The kinetics of displacement from the the immunocomplex by excess of SMZ was also studied.
