Аннотация:Abstract. The search for new regulatory factors capable of activating the antitumor defenseis today the fl agship trend in the development of immunopharmacology. The innate immunityand NK cells, as its element, play an important role in protecting against tumor and infected cells.Muramyl dipeptides (MDPs), as a structural element of the bacterial cell wall, interact with thecytoplasmic receptors of the NOD family of human cells thereby activating the innate and adoptiveimmune response. GMDP-A (glucosaminylmuramyl dipeptide) is a semisynthetic analogueof MDP, which has a number of pharmacological characteristics that can be used in the clinic forthe treatment of immunodefi ciency states, including oncology. It is known that NK cells of humanperipheral blood constitutively express NOD2 mRNA and produce its protein. The purposeof this work is to study the effect of GMDP-A on the cytolytic activity of human NK cells in vitro,as well as on the expression of gene factors involved in the implementation of the main functionsof NK cells (perforin, IFN-γ and its receptors, IL-18 receptor). While studying the effectof GMDP-A on the cytolytic activity of NK cells in relation to the HT-29 tumor cell line, a 3-foldincrease in cytolysis was observed with a ratio of target cells and effectors of 1:10. In addition, itwas shown that GMDP-A stimulates the expression of the perforin and IL-18 receptor genes anddoes not affect the level of expression of the IFN-γ genes and its receptors in NK cells of healthydonors. Thus, the ability of GMDP-A to activate the expression of the genes of perforin and IL-18receptor of NK cells is shown, which combined with a signifi cant increase in the cytolytic activityof NK cells under the infl uence of this agent, allows to consider GMDP-A as a promising agentto control the functional activity of NK cells in the treatment of cancer.Keywords: GMDP-A; muramyl dipeptide; NK cells; cytotoxicity; HT-29 cells; perforin;
