Аннотация:The kinetics of H2 oxidation by methylviologen catalyzed by hydrogenase from T. roseopersicina was studied. The initial rates of the reaction are proportional to the concentration of active sites of the enzyme and do not depend on pH or H2 concentration. The initial reaction rate is a function of oxidized methylviologen concentration according to the Michaelis mechanism. The whole kinetic curve can be linearized in Worker-Schmidt coordinates, thus suggesting the inhibiting effect of the reaction product on the rate of the reaction. The kinetic curves of H2 consumption were recorded at saturation with methylviologen. The dependences of the Km and Vmax values on the methylviologen content and pH were determined. A kinetic scheme was developed for the reaction. The values of some rate constants were calculated.
